Cancer Doctors Erupt Into Standing Ovation for "Historic" Breakthrough
The once-daily pill also improved progression-free survival and drew a nearly one-minute ovation at the ASCO meeting.
- On May 31, lead investigator Dr. Brian M. Wolpin presented trial data at the American Society of Clinical Oncology Annual Meeting showing daraxonrasib cut the risk of death by 60 percent in patients with metastatic pancreatic cancer.
- Daraxonrasib targets a mutated KRAS protein that fuels tumor growth in more than 90 percent of pancreatic cancer cases, a protein long considered "undruggable" because its smooth surface prevented drug molecules from binding.
- In the Phase 3 RASolute 302 trial of 500 patients, daraxonrasib achieved overall survival of 13.2 months versus 6.6 months for chemotherapy. Treatment discontinuation due to side effects was just 1.2 percent for daraxonrasib versus 11.2 percent for chemotherapy.
- The Food and Drug Administration granted daraxonrasib Breakthrough Therapy and Orphan Drug designations, expediting its review process. The agency is also allowing expanded access to the experimental drug for patients who meet certain criteria.
- Researchers are now testing daraxonrasib in earlier treatment lines, combination therapies, and other RAS-driven cancers including lung, colon and ovarian. ASCO chief medical officer Dr. Julie R. Gralow called the data "much more than a home run.
