Vopimetostat-Daraxonrasib Combo ‘Extremely Encouraging’ in Metastatic Pancreatic Cancer

- On Monday, Tango Therapeutics reported that its experimental drug Vopimetostat, combined with Revolution Medicines' Daraxonrasib, shrank tumors in 11 of 12 patients with previously treated pancreatic ductal adenocarcinoma.
- Investigators enrolled patients whose disease advanced after at least one treatment and who carry both a RAS mutation and a genetic deletion called MTAP, which occurs in approximately 40% of pancreatic cancers.
- Initial trial data showed 92% of patients achieved an objective response, with 90% surviving six months without disease progression, suggesting that blocking the PRMT5 enzyme provides additive benefits.
- Shares of Tango surged as much as 49% in Monday's premarket trading as Mizuho analyst Joseph Catanzaro wrote that the move into first-line treatment hints at a "potentially compelling" regimen.
- The company plans to initiate a Phase 3 trial later this year testing the combination as first-line treatment for MTAP-deleted pancreatic cancer, aiming to provide a chemotherapy-free option.
