GLP-1s may increase risk of osteoporosis and gout, new research finds
Study of over 146,000 adults links GLP-1 drugs to a 30% higher osteoporosis risk and 12% higher gout risk, with bone mineral issues also noted, researchers said.
- At the American Academy of Orthopaedic Surgeons meeting, the researchers linked GLP-1 drugs including Ozempic and Wegovy to higher osteoporosis and gout risks using five years of data from more than 146,000 adults with obesity and Type 2 diabetes.
- Investigator moved to study this after signals including a dip in obesity linked to weight‑loss drugs like Ozempic, and prior research in the Journal of Clinical Endocrinology & Metabolism, Dr. John Horneff said.
- Measured outcomes included about 4% of GLP-1 users developing osteoporosis versus over 3% of nonusers, a 30% increased risk, and gout occurring in 7.4% versus 6.6%, a 12% increase.
- The lead author said Dr. John Horneff urged more research on GLP-1s’ bone effects while clinicians recommended monitoring diet and exercise, though some opposed changing prescriptions.
- Caveats include the observational study design and its limitations, which is not yet peer-reviewed and lacks detailed medication identifiers, though analyses showed nearly double risk for bone mineral density issues and osteomalacia.
